Growth Differentiation Factor 5 Is A Paracrine Regulator In Sarcopenic Obesity

چکیده

Sarcopenic obesity (SO) differentiates itself from sarcopenia and by exhibiting a more detrimental complex condition that increases the risk of mortality in aging individuals. obese individuals suffer greater physical disability muscle weakness than sarcopenic alone. In addition, altered cellular signaling regulates increased loss SO is largely unknown. Identifying key differentially expressed genes are important for future investigation creation therapeutics strategies aimed at attenuating or preventing loss. PURPOSE: To examine global gene expression mice to determine (DE) genes. METHODS: After weaning, twenty-four were randomly assigned young (3-4 months old) aged (22-24 groups then either high-fat (HFD, 60% fat) normal chow (NC, 14% diet. RNA was isolated plantaris Next Generation sequencing bioinformatics conducted DE among experimental groups. Log2FoldChange (FC) ≥ 0.6, Log2FC ≤ -0.6 p 0.05 criteria used significant Real-Time Polymerase Chain Reaction confirm mRNA RESULTS: Aged-HFD had an ̴ 39-49% (p 0.05) reduction body mass ratio compared all other CYP1A1, RAB15, CDH22 showed main effects Age diet interacted alter GDF5 abundance. There 26-fold increase GDF-5 abundance aged-HFD CONCLUSION: associated with TGF-B SMAD signaling. The seen could be promoting collagen deposition within skeletal affecting function. These findings show may play role atrophy SO.

Growth differentiation factor 5 is a key physiological regulator of dendrite growth during development

Dendrite size and morphology are key determinants of the functional properties of neurons. Here, we show that growth differentiation factor 5 (GDF5), a member of the bone morphogenetic protein (BMP) subclass of the transforming growth factor β superfamily with a well-characterised role in limb morphogenesis, is a key regulator of the growth and elaboration of pyramidal cell dendrites in the dev...

Suppressor of cytokine signaling (SOCS)-5 is a potential negative regulator of epidermal growth factor signaling.

The suppressors of cytokine signaling (SOCS) proteins are a family of SH2 domain-containing intracellular inhibitors of cytokine signal transduction that act by several different mechanisms. Recent evidence suggests that the action of the SOCS proteins may extend beyond the cytokine receptors to signaling initiated by members of the tyrosine kinase receptor family. In this study, the ability of...

Sarcopenia and sarcopenic obesity

The aging process is associated with progressive loss of muscle mass and strength, as well as decline in physical functioning. Although consensus diagnosis has not been reached, sarcopenia is increasingly defined by both loss of muscle mass and loss of muscle function or strength. The cause of sarcopenia is suggested as multifactorial, including hormonal changes, inflammatory pathway activation...

Hepatocyte growth factor is a regulator of monocyte-macrophage function.

Hepatocyte growth factor (HGF) is a potent paracrine mediator of stromal/epithelial interactions, which is secreted as a matrix-associated inactive precursor (pro-HGF) and locally activated by tightly controlled urokinase cleavage. It induces proliferation and motility in epithelial and endothelial cells, and plays a role in physiological and pathological processes involving invasive cell growt...
